The UK Immunological Toolbox project at Pirbright has developed and validated a high throughput workflow for sequencing hundreds of hybridoma clones. The workflow employs a universal method that allows sequencing of any hybridoma clone. Just under 400 hybridomas producing antibodies against a range of host species and viral pathogens have been sequenced (cattle; 200, chicken; 100, viruses; 50 and pig;15).
The protocol used is available from the Pirbright Immunological Toolbox team to share with both internal and external collaborators. Details about this workflow will be presented at the upcoming European Veterinary Immunology Workshop. Once sequences are available this information is displayed alongside descriptive data for the antibody e.g., ITB00321. The sequences can be requested from the Immunological Toolbox team at Pirbright through the website. However, the owner maintains controls with MTA for accessing sequences if required.
This provides the UK Immunological Toolbox the capability to permanently secure these hybridoma collections for veterinary vaccinology research and presents opportunities to engineer more versatile reagents.
